Doug e Griff sono migliori amici fin dall'infanzia e sognano di girare una nuova versione del loro film preferito, «Anaconda». Spinti dalla crisi di mezza età, partono per l'Amazzonia per iniziare le riprese. Ma quando improvvisamente compare un vero anaconda gigante, il caotico e comico set cinematografico si trasforma in una trappola mortale e la loro avventura da sogno diventa una lotta per la sopravvivenza.
